Changes of Immunologically Competent Cells in the Lymph Nodes After Antigenic Stimulation in Rabbits


Abstract
Even with a large number of studies on cellular responses of immunity, the; precise nature is still uncleared. However, immune system has been characterized as foc~il aggregation of cells of mesenchymal or eadodermal origin, particulary in the spleen, lymF~h nodes, thymus, bone marrow and alimentary tract. These -cells include reticuloendothe ~,ial cells, lymphocytes, plasma ells, reticulum cells and related forms. These are usually called as the lymphoreticular of lymphoid system, but interrelationships of these cells are not Cleary defined.
In order to clarify the changes in Fiopulation of lymph node cells after antigenic stimulation and their changes in fine structures related to antibody formation, the author observed the changes in popliteai ly~~nph nodes, by light and electron microscopes, after injecting bovine gamma globulin and complete Freund.¢¥s adjuvant in the foot pad of rabbits.
Immunologically competent cells of lymphnodes initiated to proliferate in the center of secondary lymph follicles just after the antigenic stimilation, and progressively spreaded to the adjacent area occupying entire lymph node at 64 hrs after antigenic stimulation.
Most of them were large pyrorinophilic cells under light microscope. Under electron micro-scope, small lymphocytes showed increased pseudocytoplasmic process and nuclear indentation, and prominence of nucleoli; large lymphocytes showed irregularity of cytoplasmic outline and proliferation of Golgi complex; reticulum cells showed increased pseudocytoplasmic process, pinocytotic vesicles, rER, free rebosome and lysosome; lymphocytoid plasma cells, which have combined characteristics of both of lymphocyte and plasma cell, appeared at 3~40 hrs after stimulation.
The findings suggest that the proliferating cells after antigenic stimulation and initiating cells are suspected to be origenated from lymphoreticular system.
